Abnormal meiotic behavior in three species of Crotalaria PAB

The objective of this work was to compare the meiotic behavior and pollen grain viability of three species of Crotalaria. Slides for meiotic analysis were prepared by the air-drying technique. Pollen grain viability was measured by three staining procedures (Alexander's solution, tetrazolium chloride and fluorescein diacetate) and in vitro germination in a sucrose solution. Eight bivalents were observed, confirming previous reports on populations from other regions of Brazil, as well as from other countries. Application of Comet assay to assess the effects of white bean meal on DNA of human lymphocytes BJPS

This study was conducted to evaluate the potential induction of genotoxic effects of white bean flour using the Comet assay. The test was conducted with human lymphocytes present in whole blood immediately after collection, by incubation with white bean flour in three concentrations (3.92, 9.52 and 18.18 mg/mL) at 37 ºC for 4 h followed by preparation of slides. Samples were considered positive (above 20% damage) when the damage observed to cellular DNA was higher than the negative control. No genotoxic potential was found at the doses tested. However, it would be premature to suggest absence of risk to human health of DNA damage since the exposure of cells to the extract was restricted to four hours rather than a whole cell cycle. Characterization of interphase nuclei from triploid hybrids between Pennisetum purpureum Schumach and Pennisetum glaucum (L.) R. Br. Ciência e Agrotecnologia

This study evaluated the structure and the volume of interphase nuclei from root meristems of the genotypes of napier grass (Pennisetum purpureum), pearl millet (Pennisetum glaucum) and hybrids resultant of such breeding. In napier grass, nuclei were areticulate. Both pearl millet and the triploid hybrid had semi-reticulate nuclei; also, the hybrid presented a small proportion (6%) of areticulate nuclei. Pearl millet had the highest averages of nuclear dimensions, such as volume, diameter and radius, followed by the interspecific hybrid and napier grass. Citogenética de Piper hispidinervum e Piper aduncum PAB

O objetivo deste trabalho foi detectar diferenças entre duas espécies de pimenta-longa por meio de análise de cariótipos. Foram analisados cinco acessos de Piper hispidinervum (Piperaceae) C. DC. e Piper aduncum L. (Piperaceae) pertencentes à coleção de germoplasma da Embrapa Acre, utilizando-se o método de esmagamento e coloração de Feulgen. As duas espécies apresentaram número cromossômico 2n = 24 e cromossomos pequenos e metacêntricos com comprimento médio de 1,38 µm em P. hispidinervum e 1,32 µm em P. aduncum. Pelos descritores citogenéticos obtidos não há diferença entre as duas espécies.

ABSTRACT: The aim of the present study was to evaluate the effect of the colchicine and oryzalin anti-mitotic substances on the induction of tetraploid plants and foliar anatomy of two diploids clones of Solanum commersonii ssp. Nodal segments of Solanum commersonii subsp. commersonii Dun. and Solanum commersonii subsp. malmeanum Bitt. were treated with colchicine (3.5, 5.0, and 6.5mM; 72h) or oryzalin (10, 30, and 50µM; 24h). Coloração de núcleos de esporos e hifas do cogumelo Agaricus blazei Ciência e Agrotecnologia

Com o objetivo de se estudar o comportamento nuclear durante o ciclo sexual do cogumelo Agaricus blazei , técnicas de coloração e a utilização de fluorocromos foram testadas, visando a uma padronização metodológica para estudos citogenéticos futuros. Das técnicas avaliadas neste trabalho (método de Feulgen, coloração com Giemsa e fluorescência-DAPI), a coloração com Giemsa e o fluorocromo DAPI apresentaram os melhores resultados para evidenciar núcleos. O corante Giemsa permitiu a visualização de compartimentos multinucleados. Com o DAPI, não foi possível a visualização dos septos, sendo necessária a utilização adicional de calcofluor, que tem a propriedade de corar a quitina que delimita os compartimentos das hifas.

Cytogenetics of Pennisetum purpureum Schumack x Pennisetum glaucum L. hybrids and their parents Ciência e Agrotecnologia

Cytogenetic studies were carried out on five accesses of interspecific hybrids of Pennisetum purpureum and Pennisetum glaucum and their parentals, which are part of the Forage Germplasm Bank of the "Centro Nacional de Pesquisa de Gado de Leite" of EMBRAPA, Brazil. The mitotic cytogenetic analysis, performed using stain by Schiff reactive, confirmed the chromosome number of 2n = 14, 28 and 21 for the accesses of P. glaucum, P. purpureum and their hybrids, respectively. Cytotaxonomic studies were carried out on 26 accessions of Pennisetum spp. originating from the Active Germplasm Bank of Embrapa Gado de Leite-Juiz de Fora, Minas Gerais, Brazil.This study presents cytogenetics and reproductive morphological data for each one of these accessions, which allowed groupings and suggest botanical identifications to be established. Two experiments were conducted to test the allelopathic effect of sorghum (Sorghum bicolor L.) root exudates on bean (Phaseolus vulgaris L.) cell division. Research was conducted in the greenhouse of the Wistock Agricultural Research Institute of Minas Gerais State (EPAMIG) and in a laboratory of the Federal University of Lavras (UFLA). Os objetivos deste trabalho foram estabelecer um protocolo eficiente de embriogênese somática, em híbridos triploides entre capim elefante (Pennisetum purpureum Schumach.) e milheto (P. glaucum (L.) R. Br.), e avaliar por citometria de fluxo a estabilidade genômica das plantas obtidas in vitro. A embriogênese somática e a regeneração das plantas foram estabelecidas a partir de embriões zigóticos maduros de híbridos entre capim elefante e milheto. ABSTRACT: The digestibility potential of leaves from forages depends on the amount of nutrition in their tissues, with low lignin deposition in the cell wall, mainly parenchyma and phloem. This research evaluated the leaf structure of different Urochloa genotypes and discussed its potential for evaluating digestibility. The cultivars U. brizantha, U. decumbens, U. ruziziensis and three clones of U. ruziziensis (1, 95 and 97), which are under development in breeding programs, were evaluated. The objective of this work was to evaluate the genomic behavior of hybrid combinations between elephant grass (Pennisetum purpureum) and pearl millet (P. glaucum). Tetraploid (AAA'B) and pentaploid (AA'A'BB) chromosome races resulting from the backcross of the hexaploid hybrid to its parents elephant grass (A'A'BB) and pearl millet (AA) were analyzed as to chromosome number and DNA content. Genotypes of elephant grass, millet, and triploid and hexaploid induced hybrids were compared. Intra- and intergenomic chromosomal pairing in artificially polyploidized elephant grass and pearl millet hybrids PAB

Abstract: The objective of this work was to evaluate, by genomic in situ hybridization (GISH), pairing configurations as potential indicators of recombination between chromosomes of different parental genomes, in two interspecific hybrids (elephant grass x pearl millet) artificially polyploidized. Anthers from young flower buds were used in the chromosomal preparations. The genomic probe was prepared with pearl millet DNA and labeled with biotin-16-dUTP by the nick translation reaction. Blocking DNA was prepared with genomic elephant grass DNA. The homoeologous intergenomic pairing, observed in the two hybrids, indicates the possibility of recombination between chromosomes of the parental genomes.

Karyotipic asymmetry of both wild and cultivated species of Pennisetum Bragantia

This study aimed the establishment of the relation between karyotipic asymmetry values obtained for different accessions of both wild and cultivated species of Pennisetum from Germplasm Bank of Embrapa Gado de Leite/Juiz de Fora-Minas Gerais State, Brazil. Conventional cell cycle synchronization protocols and Feulgen staining method were used to obtain metaphases plates. The wild-type accessions corresponded to the species P. setosum (2n=6x=54), P. nervosum (2n=4x=36), and P. orientale (2n=4x=36), and the cultivated to P. purpureum (2n=4x=28) and P. glaucum (2n=2x=14). The variability in those species is explored in breeding programs of forages, as well as in the production of interspecific hybrids. Hybridization is a phenomenon that leads to intergenomic conflicts following the elimination of genomic sequences. In this sense, the present work aimed to study the genomic alterations occurring after interspecific hybridization of pearl millet and Napier grass with the use of cytogenetics and flow cytometry tools. The cultivated and sexually compatible species Pennisetum purpureum (elephant grass, 2n = 4x = 28) and Pennisetum glaucum (pearl millet, 2n = 2x = 14) can undergo hybridization which favors the amplification of their genetic background and the introgression of favorable alleles into breeding programs. Mitodepressive and clastogenic effects of aqueous extracts of the lichens Myelochroa lindmanii and Canoparmelia texana (Lecanorales, Parmeliaceae) on meristematic cells in plant bioassays Genet. Mol. Biol.

The cytotoxicity effect of aqueous extracts of the lichens species Myelochroa lindmanii and Canoparmelia texana (Lecanorales, Parmeliaceae) were evaluated using meristematic cells of lettuce (Lactuca sativa) and maize (Zea mays). The seasonal effect was also evaluated. Extracts of M. lindmanii and C. texana inhibited root growth and/or percentage germination, possibly due to alterations in the cell cycle. The M. lindmanii extract showed anti-mitotic effects and blocked the cell cycle in metaphase so that c-mitosis and cells with chromosome duplication were produced. The C. texana extract appeared to hinder cell division, increasing the number of interphase cells. In addition, both extracts caused an increase in percentage of cell death. Clastogenic effects...
